The Concordia Genome Foundry has a unique blend of expertise, spanning high-throughput DNA assembly, microbial engineering, and mammalian cell engineering and phenotyping. Combined with years of practical experience in laboratory automation, and a custom-built LIMS, the Genome Foundry is now well positioned to offer services to clients outside our university. Our poster provides an overview of our capabilities, along with several key case studies: 1) A 384-plasmid assembly, completed in under two weeks; 2) A 96-plasmid assembly that required maxi-prep quality DNA for iPSC transfection; 3) How our custom-built LIMS supports DNA assembly workflows; 4) Individual nucleofection of iPSC cells with 96 prime-editing plasmids, followed by direct comparisons of prime-editing efficiencies across all 96 plasmid designs. We also showcase capacities we’re building towards and include a video demonstration of our Mammalian Suite’s capabilities.
